Promoting your work effectively is a great way to spread the word, but what constitutes "effectively" is open to varied debate. It can be agreed, though, that an attractive ad or banner is a big part of it, something sleek and simple is usually best. It's a standard marketing principle, less is usually more. Another commonly agreed way to promote yourself well is to promote yourself in specific places, places where your target audience is likely to find it. Advertising a comic on a porn site, for example, would be ineffective. Advertising a comic on another comic's site, especially one like your own, would be very effective.
Finally, and I can't stress this enough, dedication to your project will overcome a lot of barriers. Producing comics consistently and posting pages often is almost absolutely necessary in becoming popular. Very few webcomics achieve notoriety without frequent updates to keep people's interest and to spark new readership. As well, being patient while your audience has a chance to grow is a virtue, helping you get through the long initial period of obscurity. Both of these are marks of dedication, commitment to seeing your project through. If you don't have this, you're already beaten.
