Something tells me you're talking about a sprite comic.
And MSPaint.
I think, what you want to do, is go up to the top, where the toolbar is, click image, scroll to the bottom of the pop-up, and click 'Draw Opaque', then, the checkmark that was once there should disappear.
If that doesn't help.
