Hey. I remember you use Paint.Net, and so do I, so here you go.
How to make an image background transparent using Paint.Net:
Copy/paste (or open) the image. I got some Kirbies from this dude to demonstrate.
Select "Magic Wand."
Click on the area outside your sprite. Dotted lines should appear around the sprite. If the dotted lines go into or mess up the sprite, press enter, lower the tolerance, and try again.
Select "Eraser."
Make the brush size large. If you have a bunch at once, this will save time. I had a whole page of Kirbies, so...
Sweep the eraser over the whole image. The sprite won't be erased - you've only selected the background. You should see a gray-and-white checked pattern behind the sprite.
When finished, press enter.
The background is now transparent.
I don't know a thing about spriting, but this IS what you wanted to know, right?

In that case, maybe you should get Paint.Net? It's free, so you don't need to do anything but download it, and it's probably more convenient to use than Paint.
You can get it here [getpaint.net] if you want it.
If you don't, I don't use Paint, so there's not a thing I can do for you! Sorry about that.

Winzip's "free" too, buddy, at least for your purposes. Download the trial for that too.

As far as I know MS Paint doesn't have transparent background capability. You're best off just using GIMP or Paint.net.
